Performance and Compatibility

Stain77 is a suitable alternative in applications where UranyLess does not provide satisfactory performance. It is particularly effective for negative staining, yielding consistent and highly reproducible contrast. In cases of excessive contrast, intensity can be reduced by floating the grid on a drop of distilled water for 1 minute following negative staining, followed by draining and drying prior to TEM examination. For positive staining of ultrathin sections, Stain77 produces contrast comparable to those obtained with UranyLess.

Protocol

• Apply a drop of Stain77 for 1-2 minutes on ultrathin sections or for negative staining of isolated particles.
• Gently drain the stain using filter paper after staining.
• For positive staining of sections: apply post-staining with lead citrate for 30–60 seconds.
• Perform a brief rinse with distilled water to finalize the process.
